1. What Glicerolo Alfasigma is and what it is used for

Glicerolo Alfasigma contains the active substance glicerolo, which belongs to the category of
laxatives—medicines used to facilitate the elimination of faeces.
This medicine is indicated for short-term treatment of occasional constipation.
Consult your doctor if you do not feel better or if you feel worse after 7 days.

2. What you need to know before using Glycerol Alfasigma

Do not use Glycerol Alfasigma

• if you are allergic to glycerol or to any of the other ingredients of this medicine (listed in section 6)
• if you have acute abdominal pain or any abdominal pain of unknown cause
• if you have nausea or vomiting
• if you have intestinal obstruction or narrowing (stenosis)
• if you have rectal bleeding of unknown cause
• if you have an acute hemorrhoidal crisis with pain and bleeding
• if you are severely dehydrated

Warnings and precautions
Consult your doctor before using Glycerol Alfasigma:

• if the need for a laxative arises from a sudden change in your previous bowel habits (frequency and characteristics of bowel movements) lasting more than two weeks, or when the use of a laxative fails to produce an effect
• if you are elderly or in poor general health
• if you frequently have difficulty passing stools or suffer from chronic constipation

Please bear in mind that:

• laxatives should be used as infrequently as possible and for no longer than seven days
• you should drink at least 6–8 glasses of water or other fluids during treatment with laxatives to help soften the stools
• it is advisable to correct dietary habits by increasing fibre and fluid intake during episodes of constipation

Abuse of laxatives (frequent or prolonged use, or excessive doses) may cause:

• persistent diarrhoea, resulting in loss of water, electrolytes, and other essential substances
• in more severe cases, dehydration or a serious decrease in blood potassium levels may occur, potentially leading to heart, nerve, or muscle dysfunction, especially if you are also taking medicines for the heart (digitalis), medicines to increase urine production (diuretics), or corticosteroids (anti-inflammatory agents)
• the need to progressively increase the dosage
• chronic constipation
• loss of normal intestinal function (decreased intestinal tone)
• abuse of laxatives, especially stimulant laxatives (contact laxatives), may lead to dependence (and therefore a possible need to progressively increase the dosage), chronic constipation, and loss of normal intestinal function (intestinal atony).

Children
In children under 12 years of age, this medicine should only be used after consulting a doctor.
Other medicines and Glycerol Alfasigma
Inform your doctor or pharmacist if you are taking, have recently taken, or might take any other medicines, including those not requiring a prescription.
Pregnancy and breastfeeding
If you are pregnant, suspect you may be pregnant, are planning a pregnancy, or are breastfeeding, consult your doctor or pharmacist before taking this medicine.
During pregnancy and breastfeeding, Glycerol Alfasigma should be used only if necessary and under direct medical supervision, after evaluating the expected benefit for the mother against the potential risk for the foetus or infant.
Driving and using machines
Glycerol Alfasigma does not impair the ability to drive or operate machinery.
However, undesirable effects may occur during treatment; therefore, it is advisable to understand your individual response to the medicine before driving or operating machinery.

3. How to use Glycerol Alfasigma

Use this medicine exactly as stated in this leaflet or as instructed by your doctor or pharmacist. If you have any doubts, consult your doctor or pharmacist.
The correct dose is the lowest dose that produces easy bowel evacuation.
Initially use the lowest recommended doses.

Use in adults and adolescents (12–18 years)
The recommended dose is 1 adult suppository as needed, up to a maximum of 1 or 2 administrations per day.

Use in children
Children aged between 2 and 11 years: 1 children's suppository as needed, up to a maximum of 1 or 2 administrations per day.

Instructions for use

Remove the suppository from its container and, if necessary, moisten it slightly to facilitate rectal insertion.
If the suppositories appear very soft, immerse the containers in cold water before opening them.
Laxatives should be used as infrequently as possible and for no longer than seven days. A diet rich in fluids enhances the effect of the medicine.

If you use more Glicerolo Alfasigma than you should
Immediately contact your doctor or go to the nearest hospital if you have accidentally taken an excessive dose of Glicerolo Alfasigma.
Cases of overdose have not been reported. However, excessive doses (laxative abuse – frequent or prolonged use or use of excessive doses) may cause:

• abdominal pain,
• persistent diarrhoea leading to loss of water, mineral salts (especially potassium), and other essential nutrients. Such losses of fluids and electrolytes should be replaced by drinking water or other liquids. Electrolyte imbalances are characterized by the following symptoms: thirst, vomiting, weakness, oedema, bone pain (osteomalacia), and hypoalbuminaemia.
• In more severe cases, dehydration or hypokalaemia may occur, which can lead to cardiac or neuromuscular dysfunction, especially if concomitant treatment with cardiac glycosides, diuretics, or corticosteroids is being administered. Abuse of laxatives, particularly stimulant (contact) laxatives, may lead to dependence (and thus possibly the need to progressively increase the dosage), chronic constipation, and loss of normal intestinal function (intestinal atony). In case of accidental ingestion/absorption of an excessive dose of glycerol, immediately contact your doctor or go to the nearest hospital.

If you stop treatment with Glicerolo Alfasigma
If you have any doubts about the use of this medicine, consult your doctor or pharmacist.

4. Possible side effects

Like all medicines, this medicine can cause side effects, although not everybody gets them. Listed below are the side effects associated with glycerol. There are insufficient data available to establish the frequency of the individual effects listed.
Side effects that may occur with the use of Glicerolo Alfasigma are:

• isolated cramp-like pains,
• intermittent abdominal pain (abdominal colic),
• diarrhoea with loss of fluids and salts; these effects are more frequent in cases of severe constipation,
• redness with burning or pain (irritation) at the rectal level.

3. How to use Glicerolo Alfasigma

Use in adults and adolescents (12 – 18 years)
The recommended dose is 1 single-dose container when needed, up to a maximum of 1 or 2 administrations per day.

Use in children
Children aged 6 to 11 years: 1-2 single-dose containers for children when needed, up to a maximum of 1 or 2 administrations per day.
Children aged 2 to 6 years: 1 single-dose container for children when needed, up to a maximum of 1 or 2 administrations per day.

Instructions for use

1. Remove the cannula cap by bending it at the base (as shown in the figure).

2. Place your index finger and thumb on the round ring located above the bulb, and with the other hand, bend the cannula cap until it detaches from the body of the container. During this operation, never grasp the bulb, otherwise the medicine may be expelled before use.

3. It may be helpful to lubricate the cannula with a drop of the solution itself before inserting it into the rectum and compressing the bulb.

4. Remove the cannula while keeping the bulb compressed.

Use the container for a single administration only; discard any remaining medicine.
Laxatives should be used as infrequently as possible and for no more than seven days. A diet rich in fluids helps enhance the effect of the medicine.
